The bladder epithelium consists of urothelial cells that play a vital role in maintaining a strong barrier against bacterial invasion. However, these cells are highly variable between individual cultures and are not consistently available. The spontaneously immortalised cell line HBLAK\u00a0derived from a primary culture of uroepithelial cells was first characterised\u00a0by\u00a0<a href=\"https:\/\/doi.org\/10.3233%2FBLC-160065\" target=\"_blank\" rel=\"noreferrer noopener\">Hoffmann et al.<\/a>\u00a0HBLAK has become a more conveniently available model for urological research than primary urothelial cultures.<\/p>\n\n\n\n<p>The highlights of CELLnTEC\u2019s human bladder long-term epithelial cells HBLAK include:<\/p>\n\n\n\n<ul class=\"wp-block-list\" style=\"font-size:16px\">\n<li>convenience of long-term cell growth without senescence<\/li>\n\n\n\n<li>at least six months growth post-delivery<\/li>\n\n\n\n<li>supplied at approx. passage 10<\/li>\n\n\n\n<li>cryopreserved in a fully-defined medium<\/li>\n\n\n\n<li>ability to complete 2D\/3D differentiation and organoid formation<\/li>\n<\/ul>\n\n\n\n<h3 class=\"wp-block-heading\"><strong>Applications of HBLAK:<\/strong><\/h3>\n\n\n\n<p><a href=\"https:\/\/www.mdpi.com\/2076-0817\/12\/4\/523\" target=\"_blank\" rel=\"noreferrer noopener\">Wu et al.<\/a>&nbsp;and&nbsp;<a href=\"https:\/\/www.mdpi.com\/2072-6694\/12\/4\/1023\" target=\"_blank\" rel=\"noreferrer noopener\">Lang et al.<\/a>&nbsp;cultured&nbsp;<strong>HBLAK<\/strong>&nbsp;in the&nbsp;<strong>CnT-PR&nbsp;<\/strong>medium that establishes as many proliferative progenitors as possible in culture. They further used the&nbsp;<strong>CnT-PR-D<\/strong>&nbsp;medium&nbsp;for 2D differentiation on&nbsp;<strong>HBLAK<\/strong>.<br><br>Most of the available biomimetic<em>&nbsp;in vitro<\/em>&nbsp;models of the human urothelium show a short lifespan in a urine environment.&nbsp;<a href=\"https:\/\/www.frontiersin.org\/articles\/10.3389\/fcimb.2023.1128132\/full\" target=\"_blank\" rel=\"noreferrer noopener\">Jafari et al.<\/a><strong>&nbsp;<\/strong>established human 3D bladder cultures with&nbsp;<strong>HBLAK<\/strong>,&nbsp;<strong>CnT-PR<\/strong>, and&nbsp;<strong>CnT-PR-3D<\/strong>. They reported an advanced barrier-forming, 100% urine-tolerant, 3D urothelial microtissue model that can be used to elucidate UTI host-pathogen interactions.<br><br><a href=\"https:\/\/www.sciencedirect.com\/science\/article\/abs\/pii\/S0168365920304892?via%3Dihub\" target=\"_blank\" rel=\"noreferrer noopener\">Lau et al.<\/a>&nbsp;cultured&nbsp;<strong>HBLAK<\/strong>&nbsp;cells in&nbsp;<strong>CnT-PR&nbsp;<\/strong>and subsequently used the&nbsp;<strong>CnT-PR-3D&nbsp;<\/strong>medium to create 3D stratified human bladder organoids.
