In the absence of R-spondin, the Frizzled receptor (Fz) is internalised through ubiquitination mediated by the ubiquitin ligase RNF43\/ZNRF3. This leads to a decrease in Fz on the cell membrane and insufficient activation of Wnt signals.<\/p>\n\n\n\n<p>On the other hand, when R-spondin is present, RNF43\/ZNRF3 forms trimers with LGR and R-spondin, leading to its endocytosis.
