Colony Formation Assays
Hematopoietic Stem Cell (HSC) potential is routinely measured through myeloid colony forming units (CFU) in a semi-solid culture environment. In the presence of cytokines, unipotent progenitor cells are capable of giving rise to colonies with granulocytes (CFU-G), macrophages (CFU-M), erythrocytes (BFU-E) and megakaryocytes (CFU-Mk) while stem/progenitor cells with multipotency give rise to colonies with multiple lineages (CFU-GM and CFU-GEMM). Long term culture-initiating cell (LTC-IC) assays are used to identify the most primitive HSC which is capable of maintaining its multipotency after long term culture.
Expansion and Differentiation of Stem and Progenitor Cells
Mesenchymal Stem Cells/Multipotent Stromal Cells (MSC) are isolated by AllCells from multiple sources, including bone marrow, cord blood, adipose tissue and dental pulp from deciduous teeth. MSCs have been shown to possess multilineage differentiation potential, including but not limited to, adipocytes, chondrocytes, osteoblasts, neurons and myocytes. AllCells offers custom services for the quantification of MSC using colony forming fibroblast (CFU-F) assays and characterization of MSC derived adipocytes, chondrocytes and osteoblasts.
Custom Differentiation of Stem and Progenitor Cells
In addition to characterization of HSC and MSC potential, AllCells offers services in custom differentiation of our stem and progenitor cells. AllCells can provide our customers with populations of difficult to isolate cells which have been expanded and differentiated in culture, including megakaryocytes, dendritic cells, mast cells, and early erythroid cells.
The Bioservices team can provide customized applications of the system to accelerate your research. In addition to stem cell assays they also provide a number of other services including: immunoassays, flow cytometry, protein analysis, genotyping and tissue culture services.
