Protein G is a well-established ligand for affinity chromatography resins used for isolation and characterization of immunoglobulins. Protein G binds to the Fc region of mammalian IgGs thus exhibiting high affinity for many IgG subclasses including human, rabbit, mouse, rat, sheep, pig, horse, cow or goat. In contrast to protein A, protein G binds all human and mouse IgG subclasses, including human IgG3 and mouse IgG1 as well as all rat IgG subclasses. Furthermore, protein G also binds to CH1 portion of the Fab region making this ligand well-suited for the purification of more complex constructs such as bispecific antibodies.

Ion Exchange Chromatography (IEX) is one of the most frequently used techniques for biotherapeutics separation since it separates biomolecules based on their net surface charge. For oligonucleotide purification, anion exchange chromatography (AEX) is an effective technique that can be helpful for the removal of incomplete or erroneous sequences, resulting in high purity and yield. However, there is always a need to improve the purity and yield of the purification process with a new purification matrix. In this study, two new anion-exchange agarose resins were developed by Agarose Bead Technologies (ABT), Q10F Rapid RunTM High Sub (QRR10FPT1) and QRRPT1 Rapid RunTM High Sub Agarose Bead Fine (QRRFPT1), which greatly improved the loading capacity, purification yield, and purity of a crude mRNAs oligonucleotide.
