Description:
This novel human mast cell line is spontaneously derived from non-transformed hematopoietic progenitor cells cultured with stem cell factor and interleukins. Cells are characterized by their ability to survive and proliferate without the addition of growth factors and display metachromatic cytoplasmic granules while expressing a range of mast cell-related genes and proteins, including tryptase, FcεRI, and c-kit. They respond to FcεRI cross-linkage by releasing mediators such as β-hexosaminidase and prostaglandin D2. Importantly, these cells do not require stem cell factor for maintenance, exhibit high levels of normally signaling c-kit and FcεRI, and are valuable for functional human mast cell studies due to their stable phenotype and functional characteristics.Note: A difference in surface FcεRI expression, compared to the data in the original publication, has been observed by the depositor of the LUVA cells. Approximately 5% FcεRI expression by cell surface staining has been reported. However, intracellular staining and mRNA expression for FcεRI shows high and very high levels, respectively. The reason for this difference is not clear.

Growth Conditions:
Upright PriCoat™ T25 Flasks (G299) are recommended for optimal cell culture. PriGrow X Series Medium (TM0843) + 2 mM L-glutamine (G275) + 1% Penicillin/Streptomycin Solution (G255), 37.0°C, 5% CO2.Note: Grow cells in upright culture vessels. Do not culture/freeze-down cells with FBS.
