Background:
L6 myoblasts were originally isolated by Yaffe from the first two generations of primary cultures of rat thigh muscles in the presence of methylcholanthrene. In culture, L6 cells can fuse to form multinucleated myotubes and striated muscle fibers. The extent of L6 cell fusion decreases with increasing passage number; therefore, L6 cells should be frozen at early passages and periodically recloned to select for cells with strong fusion ability. If the cells completely cover the culture dish, the myogenic component of L6 cells will be rapidly depleted.
