Real Time PCR sequence-specific probes with reporter and quencher; probe is cleaved by Taq polymerase during amplification, separating dye from quencher. Real-time PCR monitors the accumulation of PCR product (amplicon) cycle-by-cycle using a fluorescent signal that increases proportionally with the amount of amplified DNA.
