Background:
TNFRSF7, also known as CD27, is a type I membrane glycoprotein of the TNF receptor family, with a molecular weight of 55KDa, forming disulfide-linked homodimers. Its extracellular domain contains three cysteine-rich repeats, and it binds to its trimeric ligand CD70 (CD27L). It is expressed on T-cells, NK-cells and B-cells, mediating antigen-specific T-cell activation, expansion and memory formation. CD70 ligation activates the TRAF2-PTPN6 axis, inhibiting LCK phosphorylation to regulate T-cell memory adaptation. It also modulates hematopoiesis via negative feedback on leukocyte differentiation and regulates Th2 lymphocyte function. Soluble CD27 (sCD27), its extracellular domain, is released post lymphocyte activation and serves as a diagnostic marker in certain diseases.

Test Principle:
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse TNFRSF7. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse TNFRSF7 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse TNFRSF7, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ? 6 nm. The OD value is proportional to the concentration of Mouse TNFRSF7. You can calculate the concentration of Mouse TNFRSF7 in the samples by comparing the OD of the samples to the standard curve.
