Background:
Dipeptidyl Peptidase IV (DPPIV, also CD26 and adenosine deaminase complexing protein 2) is a serine protease that releases Xaa-Pro dipeptides from the N-terminus of oligo- and polypeptides. As a type II membrane protein, it includes a small cytoplasmic tail, a transmembrane region, and a large extracellular domain with glycosylation sites, a cysteine-rich region, and a Ser-Asp-His catalytic active site. Native DPPIV exists as a non-covalently linked homodimer on cell surfaces, with its soluble form (intact extracellular domain) present in human serum and other body fluids.
DPPIV plays a key role in multiple physiological and pathological processes: it interacts with ADA and CD45 to co-stimulate the CD3/T-cell receptor complex, cleaves N-terminal Xaa-Pro-containing chemokines, altering their receptor specificity and function, degrades peptide hormones (e.g., glucagon-like peptide-1) to reduce bioactivity, with DPPIV inhibitors for type 2 diabetes in late-stage clinical trials, truncates procalcitonin (a marker for systemic bacterial/fungal infections), and interacts with HIV-1 Tat protein, whose ADA binding is inhibited by HIV envelope protein gp120.

Test Principle:
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human DPP4. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human DPP4 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human DPP4, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ? 15 nm. The OD value is proportional to the concentration of Human DPP4. You can calculate the concentration of Human DPP4 in the samples by comparing the OD of the samples to the standard curve.
