MC38 cells come from the C57BL/6 mouse colon adenocarcinoma cell line and are widely used as a mouse model for colorectal cancer research. This cell line has a high mutation rate, especially in terms of its mutanome and neoantigen expression, making it highly sensitive to immune checkpoint inhibitor therapies. Their response to endogenous CD8 T cell attacks on neoantigens highlights their value in studying immune interactions within the tumor environment, positioning the MC38 model as a key immunoreactive mouse tumor model. MC38 cells can form tumors and metastases in syngeneic C57BL/6 mouse hosts or immunodeficient mice. The MC38 colon adenocarcinoma model, used especially in orthotopic mouse studies, is recognized for its immune responsiveness, making it an effective platform to evaluate immunotherapies, including radiotherapy, checkpoint inhibitors, and other novel treatments. MC38 cells express colon markers like claudin-1 and SATB2, which is crucial for studying the genomic and epigenomic basis of colorectal adenocarcinoma and identifying potential therapeutic targets. The immunological characteristics of the MC38 xenograft model make it a versatile tool for cancer research, particularly in the context of colorectal adenocarcinoma. With its high mutanome and neoantigen load, the MC38 colon adenocarcinoma model serves as a classic immunoreactive mouse model, facilitating exploration of the complex dynamics between colorectal tumor cells and the host immune system.
