Hepa1-6-GFP is a cell line derived from BW7756 hepatoma induced in a C57L mouse. It expresses AFP, α1-antitrypsin, and amylase, and tests negative for ectromelia virus. The cells can be propagated in serum-free medium consisting of 75% DMEM and 25% Waymouth?s MAB 87/3, supplemented with 3x10^-8 M selenium. For drug screening: Fluorescent gene fragments are integrated into the cellular genome via lentiviral infection, enabling the cells to express fluorescent proteins that can be visualized under a fluorescence microscope, allowing easy tracking and detection of labeled cells. Owing to variable fluorescence expression resulting from lentiviral transduction, antibiotic selection may be used to enrich for cells with high fluorescent protein expression. During routine culture, supplemental selection is recommended at a final concentration of 4 ug/mL puromycin, applied 2?3 times per month or at a customized frequency. A single round of supplemental selection is also advised following thawing of cryopreserved cells. Daily addition of puromycin during routine culture is not required.
