Hepa1-6-LUC is a cell line derived from BW7756 hepatoma induced in a C57L mouse. It expresses AFP, α1-antitrypsin, and amylase, and tests negative for ectromelia virus. The cells can be propagated in serum-free medium consisting of 75% DMEM and 25% Waymouth?s MAB 87/3, supplemented with 3x10^-8 M selenium. For drug screening and reporter assays: The luciferase (Luc) gene fragment is integrated into the cellular genome via lentiviral infection, enabling the cells to express luciferase. This cell line is commonly used to establish various subcutaneous, orthotopic, or metastatic CDX tumor models, as well as for in vivo bioluminescence imaging and promoter activity analysis. Owing to variable luciferase expression associated with lentiviral transduction, antibiotic selection can be performed to enrich positive cells. The culture conditions for the luciferase-labeled line are identical to those for the wild-type cells. For continuously cultured cells, selection is recommended every 1?2 months. For selection, puromycin is added directly to the culture medium at a final concentration of 3.5 ug/mL, and cells are cultured and passaged normally for one week. For long-term cryopreserved cells, selection can be performed in the second passage after thawing once the cells have stabilized, to maintain the proportion of positive cells. It is recommended not to use the cells for experiments during the selection period, as antibiotics may interfere with the results of certain assays.
