The 4T1 cell line is a 6-thioguanine-resistant subline selected without mutagenesis from the 410.4 tumor line. When injected into BALB/c mice, 4T1 cells spontaneously form highly metastatic tumors that spread to the lungs, liver, lymph nodes, and brain, while simultaneously forming a primary tumor at the injection site. Metastasis induction does not require removal of the primary tumor. The growth and metastatic characteristics of 4T1 cells in BALB/c mice closely resemble those of human breast cancer, making this model representative of human stage IV breast cancer. The kinetics of metastasis induced by 4T1 cells are similar in both postsurgical and nonsurgical settings, allowing the model to be used as either a postsurgical or nonsurgical system. Compared with other tumor models, the intrinsic 6-thioguanine resistance of 4T1 cells enables detection of even tiny metastatic cell clusters (as few as a single cell) in many distant organs, eliminating the need for lymph node counting or organ weight measurement. The 4T1-GFP cell line was constructed from 4T1 cells via PiggyBac (PB) transposon transfection. It is a stable cell line capable of long-term, high-level GFP expression. This cell line features low passage number, high viability, and good condition, making it suitable for assays including flow cytometry and fluorescence imaging.
