Background:
The CT26 cell line is an undifferentiated colon carcinoma cell line induced by N-nitroso-N-methylurethane. A cloned derivative was designated CT26.WT. The lethal subclone CT26.CL25 was generated by stable transfection of CT26.WT with the retroviral vector LXSN carrying the lacZ gene, which encodes the tumor-associated antigen (TAA) β-galactosidase (β-gal). Though CT26.CL25 constitutively expresses β-galactosidase, these cells exhibit largely consistent growth rates and in vivo lethality with CT26.WT in immunocompetent mice. CT26.WT and CT26.CL25 are widely used in paired models for evaluating immunotherapeutic approaches and investigating host anti-tumor immune responses. The CT26.WT-GFP cell line was constructed from CT26.WT cells by lentiviral transduction and is a stable cell line capable of long-term, high-level GFP expression. This cell line exhibits low passage, high viability, and good condition, making it suitable for assays such as flow cytometry and fluorescence imaging. Selection drug concentration: Puro = 10.0 ug/mL.

Tumorigenic:
Yes, in BALB/c mice. Mice inoculated, subcutaneously, developed lethal tumors at 80% frequency with 1x10^3cells and at 100% with 1x10^4 cells. Pulmonary metastases developed when mice were inoculated, intravenously, with 1x10^4 cells.
