The MC38 cell line is a murine colon adenocarcinoma cell line derived from C57BL/6 mice and widely utilized as a preclinical model for colorectal cancer research. This cell line presents a high mutation burden with abundant mutanome and neoantigen load, conferring high sensitivity to immune checkpoint inhibitor therapy. Its susceptibility to endogenous CD8+ T-cell-mediated neoantigen targeting underscores its value for investigating immune crosstalk in the tumor microenvironment, making MC38 a core immunoresponsive syngeneic tumor model. MC38 cells efficiently form primary tumors and distant metastases in both syngeneic C57BL/6 and immunocompromised mice. The orthotopic MC38 model is well-recognized for robust immunogenicity, serving as a reliable platform to evaluate checkpoint inhibitors, radiotherapy and other novel anti-tumor agents. It constitutively expresses colon-specific markers including claudin-1 and SATB2, which support mechanistic research on the genomic and epigenomic landscape of colorectal adenocarcinoma and the development of targeted therapies. Owing to its high mutational and neoantigen burden, the MC38 model is an ideal murine system to dissect the dynamic interactions between colorectal tumor cells and host immunity. The MC38-GFP cell line was constructed from MC38 cells by lentiviral transduction and is a stable cell line capable of long-term, high-level GFP expression. This cell line exhibits low passage, high viability, and good condition, making it suitable for assays such as flow cytometry and fluorescence imaging. Selection drug concentration: Puro = 4.0 ug/mL.
