The Panc02 cell line is a widely used murine model for studying pancreatic ductal adenocarcinoma (PDAC), the most common and aggressive subtype of pancreatic cancer. Panc02 cells were originally derived from a chemically induced pancreatic tumor in C57BL/6 mice. This cell line is highly valuable for preclinical research, as it can be implanted orthotopically in syngeneic mice to recapitulate the native tumor microenvironment and facilitate research into PDAC immune responses and therapeutic resistance mechanisms. Studies utilizing Panc02 have advanced our understanding of the immunosuppressive microenvironment in PDAC. Panc02 tumors are heavily infiltrated by regulatory T cells (Tregs), which inhibit antitumor immune activity. Low-dose gemcitabine has been shown to selectively deplete Tregs in Panc02 tumor-bearing mice, thereby enhancing antitumor immunity and moderately prolonging survival. These findings indicate that immunomodulatory intervention is a promising therapeutic strategy for PDAC. Beyond immunotherapy research, Panc02 is also applied in studies of necroptosis, a regulated form of programmed cell death. Inhibition of Aurora Kinase A in Panc02 cells induces necroptosis, a key mechanism for overcoming apoptosis resistance in PDAC. This offers a potential therapeutic direction for targeting apoptosis-resistant tumor cells via activation of non-apoptotic cell death pathways. The Panc02-GFP cell line was constructed from Panc02 cells by lentiviral transduction and is a stable cell line capable of long-term, high-level GFP expression. This cell line exhibits low passage, high viability, and robust cellular status, making it suitable for assays such as flow cytometry and fluorescence imaging. Selection drug concentration: Puro = 4.0 ug/mL.
