Background:
RWPE-1 cells were established by transforming epithelial cells from the peripheral zone of a normal adult human prostate tissue section with human papillomavirus type 18 (HPV-18). When cultured in three-dimensional Matrigel, RWPE-1 cells form acini and secrete PSA into the culture medium in response to androgens [PubMed:11170142]. RWPE-1 cells also form acini and produce PSA when injected subcutaneously into male nude mice in combination with Matrigel or stromal cells [PubMed:11304724]. RWPE-1 cells further transfected with the Ki-ras gene using the Kirsten murine sarcoma virus gave rise to the tumorigenic RWPE-2 and RWPE2-W99 cell lines [PubMed:9214605]. Additionally, treatment of RWPE-1 cells with N-methyl-N-nitrosourea (MNU) generated a series of tumorigenic cell lines that model different stages of prostate cancer progression, namely WPE1-NA22, WPE1-NB14, WPE1-NB11, and WPE1-NB26. According to the provider, RWPE-1 cells have tested negative for hepatitis B, hepatitis C, and human immunodeficiency virus.
