Background:
The 12Z cell line was derived from an endometrial peritoneal biopsy specimen from a 37-year-old woman. It was immortalized by transfection with a plasmid encoding the SV40 large T antigen (SV40T). The cell line expresses pan-cytokeratin and N-cadherin. Retrograde menstruation, which enables endometrial tissue to implant outside the uterus, is one of the most widely studied mechanisms underlying the pathogenesis of endometriosis. The invasive capacity of endometrial cells is considered critical in this process. The immortalized human endometrial epithelial cell line 12Z represents a well-established model for endometriosis, as it was derived from N-cadherin?positive endometriotic tissue, with N-cadherin positivity serving as a marker of invasiveness. This renders the 12Z line a suitable model for investigating disease progression and therapeutic development. The 12Z cell line is commonly used in 2D and 3D invasion assays as well as biochemical assays to study the pathophysiology of endometriosis.
