Background:
The U-2 OS cell line (originally named 2T) was established in 1964 by J. Pont?n and E. Saksela from a moderately differentiated sarcoma of the tibia in a 15-year-old female. When co-cultured with WI-38 cells or tested by complement fixation (CF) assay for simian virus 40 (SV40), respiratory syncytial virus (RSV), or adenoviruses, no viruses were detected. U-2 OS cells express insulin-like growth factor I (IGF-I) and II (IGF-II) receptors, as well as osteosarcoma-derived growth factor (ODGF). U-2 OS-LUC-EGFP is a derivative cell line generated by infecting parental U-2 OS cells with a recombinant lentivirus carrying luciferase and EGFP genes. This cell line stably expresses both luciferase and enhanced green fluorescent protein (EGFP), enabling its application in various in vivo and in vitro experiments requiring cell tracking. Selection drug concentration: Puro = 2.0 ug/mL.
