Cas12a recognizes a T-rich PAM sequence (specifically 5'-TTTV-3') located at the 5' end of the target site. It employs a single RuvC domain to generate staggered "sticky" ends, facilitating more efficient homology-directed repair (HDR).Cas12a mRNA is is produced through in vitro transcription, featuring a Cap1 structure and a Poly(A) tail.
