Defined Immune Response Tracking in Mice: OVA MHC Tetramers

T cell-dependent OVA (ovalbumin) antigen is commonly used as a model protein for studying antigen-specific immune responses in mice. SIINFEKL is one peptide sequence of the OVA used to precisely probe various aspects of antigen presentation, including the kinetics of peptide generation, MHC class I surface stability, and presentation efficiency following pharmacological and genetic manipulations including high throughput drug screening. In addition, the SIINFEKL peptide in the presence of an adjuvant is used in mice models to determine the capacity to raise an immune response. Primary immunisation with the peptide may lead to memory T cell responses that generate strong recall responses which can be detected by staining with SIINFEKL (OVA)-tetramer. The percentage of OVA-specific CD8+ T cells indicates to the researchers the percentages of antigen-specific T cells and their effector phenotypes. The results would demonstrate the success of the vaccine against infectious diseases or cancer (1-4).
The H-2Kb OVA Tetramer detects OVA-specific CD8 T cells in C57BL/6 and other mouse strains expressing the class I allele, H-2Kb. Whereas I-Ab OVA-Specific detects CD4 T cells in I-A b mouse strains expressing the class II allele.
