SARS-Cov-2 Neutralising Antibody Kits

When someone contracts COVID-19, the host’s immune response can be measured by the presence of neutralising antibodies. Neutralising antibodies prevent the interaction between SARS-CoV-2 virus and receptors on human cells, giving the patient immunity.

Neutralising antibody detection is therefore very important in understanding and treating COVID-19. For example, to:

  1. Test neutralising antibody induction during vaccine development, and how long antibodies are present for after infection
  2. Confirm COVID-19 patient immunity
  3. Allow donor selection in convalescent plasma therapy (using the antibodies of immune patients to help patients with active COVID-19)
  4. Identify compounds that can interfere with the mechanism of viral infection, to prevent or treat COVID-19

COVID-19 Mechanism of Infection: RBD-ACE2 Complex

The mechanism of COVID-19 infection works through the interaction between the SARS-CoV-2 Spike protein, and the ACE2 receptor on the surface of human cells. The receptor binding domain (RBD) of the Spike protein binds to ACE2 receptors, allowing the virus to infect cells and to self-replicate.

Neutralising antibodies bind to the RBD on the Spike protein, preventing the virus from binding to ACE2 receptors and infecting cells.

Neutralising Antibody Detection

Neutralising antibody detection kits work on the ELISA principle, normally, the reaction between Horseradish peroxidase (HRP) and colourmetric substrate. To detect Neutralising antibodies, either ACE2 or RBD is labelled with HRP, and the other is bound to a plate. In the example below, ACE2 is labelled.

If the donor sample contains neutralising antibodies, RBD and ACE2 will not bind together. ACE2 will be washed off, and there will be no signal

If there are no neutralising antibodies, RBD and ACE2 will bind and ACE2 will not be washed off. The HRP on ACE2 will then be able to react with the colourmetric substrate, producing a signal.
